1. (a). What is biotechnology?

(b). Define the following terms:

i) A Construct

ii) Cloning

iii) Restriction enzymes

iv) Transformation and

v) Recombinant DNA.

2. Write briefly on the following:

(a). Nitrogen fixation

(b). Herbicide resistance and

(c). Random Amplification of Polymorphic DNA.

(d). List three methods by which a plant can convey herbicide resistance.

3. (a). Write briefly on the following:

(i) Sterile insect release method (SIRM).

(ii) Role of nitrogen in the biosphere.

(iii) Genetically modifying insect pests of crops as a control tactic

(b). What is recombinant DNA?

(c). List three herbicides and three herbicide-tolerant plants that have been developed for each of the three herbicides.

4 (a).Why are transgenic crops needed?

(b). What are diazotrophs?

(c). What is a genetic marker?

(d). List three commonly used genetic markers.

(e). Give five precautions that should be taken in order to get good response to biofertilizer application.

5 (a). Discuss the three major genetic traits that are used in breeding for resistance to fungal diseases in crops.

(b). Bt Toxin is a toxin encoded by the bacterium named Bacillus thuringiensis. Explain briefly the principle behind its mode of action and the target insects.

6. Discuss briefly the basic strategy for transforming a plant starting from assemblage of the construct.

7. (a). How does non-lethal mutation differ from neutral mutation?

(b).Discuss briefly the stages of micropropagation.
